Preliminary data suggest that HR landscape promotes metabolic reprogramming to sustain high-proliferation rates accompanied by a higher oxidative state compared with standard risk disease. Given these premises, this project aims to explore the potential benefit of metabolism-targeted therapy in HR-MM patients. For this purpose, we plan:
1) to evaluate multiomics (including genomics, transcriptomics, proteomics) and the resulting metabolic profile of HR-MM.
2) to study the proteomic and the resulting metabolic profile of primary MM cells obtained by HR-MM patients, performing different culture conditions in order to understand how their metabolism, signaling, cell growth and survival are influenced by the availability of carbon sources.
3) to screen a “compound library” and/or specific dietetic regimen (i.e. isocaloric ketogenic diet) to identify novel molecules targeting metabolic vulnerabilities
